{"created":"2023-05-15T10:54:59.574339+00:00","id":4744,"links":{},"metadata":{"_buckets":{"deposit":"4611ff2e-0ca7-4e98-b415-f2980d7fec61"},"_deposit":{"created_by":17,"id":"4744","owners":[17],"pid":{"revision_id":0,"type":"depid","value":"4744"},"status":"published"},"_oai":{"id":"oai:juen.repo.nii.ac.jp:00004744","sets":["993:994:1005"]},"author_link":["14520","14521","14522","14523","14524","14525","14526","14527"],"item_10002_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1999-09","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"109","bibliographicPageStart":"99","bibliographicVolumeNumber":"19","bibliographic_titles":[{"bibliographic_title":"上越教育大学研究紀要"}]}]},"item_10002_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"プランクトン(浮遊生物)は,生物の多様性を学習する教材として大学の実験・観察の授業にしばしばとりあげられている。その多くの授業では光学顕微鏡観察が主体であり,微細形態の観察・比較ができる走査型電子顕微鏡は殆ど活用されていない。最近,低真空走査型電子顕微鏡を用いたプランクトン観察のための新しい凍結乾燥法が提案された。この方法は,従来の方法に比較して短時間でプランクトンの観察試料が作製でき,その微細形態もよく保持されると報告されている。プランクトンの観察授業で走査型電子顕微鏡を活用するためには,試料が短時間で作製できると共に,体制や大きさが多様なプランクトンの多くの種で微細形態が保持される必要がある。そこで本研究では,その活用のための基礎的研究として,野外で採集した淡水プランクトンを中心に,低真空走査型電子顕微鏡を用いて凍結乾燥試料を作製し,その微細形態が保持されるかどうかを検討した。自然乾燥した場合,ミカヅキモやミドリムシ等の柔らかいプランクトンは著しく変形した。凍結乾燥した試料では,比較的柔らかいプランクトンの多く(渦鞭毛藻類,緑藻類,緑虫類,など)の種で,臨界点乾燥試料と同様に,微細形態がよく保持されていた。また,輪虫類の口部繊毛環も比較的よく保存されていた。しかし,緑虫類では鞭毛を欠く細胞が高頻度で出現した。グルタルアルデヒドとオスミウム酸で二重固定した試料でも,結果は改善されなかった。今回の実験・観察から,低真空走査型電子顕微鏡を用いた凍結乾燥法は,緑虫類の鞭毛観察では配慮を要するが,多くの淡水プランクトンの微細形態観察には有効であることが示唆された。プランクトン観察授業における低真空走査型電子顕微鏡の活用について考察する。","subitem_description_type":"Abstract"},{"subitem_description":"The plankton has been used as a material to teach the diversity of organisms in classes of Biology in university. In these classes, the light microscope has been exclusively employed in observing plankton, and, so far as we know, the scanning electron microscope (SEM) has not been applied to it. Recently, a new preparation method of plankton for SEM observation was proposed, in which plankton having been immersed in distilled water was frozen with liquid nitrogen and then the water was sublimated within a specimen chamber of a low-vacuum SEM (LV SEM). The application of this method is reported to enable to prepare specimens easily. For the application of SEM to the classes for plankton observation, not only the convenience in specimen preparation but also the preservation of surface microstructures of many species of plankton varying in organization and size would be required. The aim of the present investigation is primarily to examine the preservation of microstructures of various kinds of plankton prepared by the new freeze-drying method. When dried under natural conditions, soft plankton, such as Closterium and Euglena, showed remarkable deformation. By contrast, soft planktonic organisms (some species of the Chlorophyta, Dinophyta, Euglenophyta, etc.) prepared by the freeze-drying method, like those prepared by the critical-point drying method, maintained their surface microstructures. The ciliary bands of rotifers were preserved, but many of euglenoid cells lacked their flagella. Treatment of euglenoids with glutaraldehyde and osmium tetroxide prior to freeze-drying did not improve the maintenance of their flagella. The present examination suggests that the freeze-drying method using LV SEM may be effective in preservation of microstructures of various kinds of soft planktonic organisms, except for euglenoid flagella.
